Digests and ladder are ready-to-run. The 80 μL volume is enough for 6 separate
wells or about 13 μL per well. If necessary, 72 μL of glycerin may be added to each
microcentrifuge tube. Once blended, 25 μL of the resulting solution can be added to
each well. Ready-to-run DNA should be stored in the freezer. Available September

Prepare Electrophoresis
Buffers
The following recipes will enable you to prepare your own buffer solutions for
nucleic acid (RNA and DNA) electrophoresis. These buffer solutions are used to fill
the electrophoresis chamber and also to prepare the agarose gel mixture. Recipes
are for 10X concentrate solutions—offering longer shelf life and requiring less space.
Mix the ingredients detailed below with 750 mL of distilled or deionized water. Stir to
dissolve, and dilute to a final volume of 1 liter. Prepare the 1X working strength solution,
as needed, by mixing 1 part concentrate with 9 parts distilled or deionized water.
10X Tris-Borate-EDTA (TBE) Buffer

Convenient and trouble-free. Simply melt in a hot water bath or microwave, cool to

Prepare an Agarose Gel
The standard concentration of agarose in the gel is 0.8%—a concentration that offers a
compromise between band resolution and running time. The following directions are
for 100 mL of an 0.8% agarose solution.
1. Stir 0.8 g of agarose into 100 mL of working strength (1X) electrophoresis buffer (TBE
or TAE) in a glass Erlenmeyer flask. Stopper with non-absorbent cotton, or foam
plug.
2. Dissolve agarose by heating in a microwave (30–40 seconds, stir, repeat) or on a
hot plate. Heat until solution is clear and agarose appears to be fully dissolved. Stir
frequently and do not allow solution to boil for more than a few seconds.
3. Prepare the casting tray, place the well comb, and pour the gel(s) when the agarose
solution has cooled to approximately 60 °C.
4. Allow the gel to fully solidify on a flat, level surface for 20 to 30 minutes. Gel should
be opaque and firm to the touch.
Gel Loading Solution
Combine the gel loading Solution with DNA digests to both visualize the sample run
and to make the sample sink into the well. This dense, buffered solution contains
two dyes. Bromphenol blue migrates at the same rate as a 200–400 base pair DNA
fragment, at the beginning of a typical run. Xylene cyanole migrates at the same rate
as a 4,000 base pair DNA fragment and helps visualize the middle to end. Usually add
1 μL of the gel loading Solution for every 5 μL of digest.

Methylene Blue Electrophoresis Stain
Methylene blue electrophoresis staining solution interacts with both RNA and DNA—
and does not require ultraviolet light for visualization. Bands will appear dark blue
on a light blue background. This bottle contains 300 mL of 10X concentrate and will
make three liters of working solution.

Reagents for Protein Electrophoresis
Tris-Glycine-SDS Buffer is optimized for separation of proteins (as opposed to
nucleic acids) during agarose gel electrophoresis. The Protein Staining Solution
binds to protein molecules so the protein bands will be visible in the gel.
